- Lige Leng of Xiamen University and colleagues reported in PLOS Biology (March 2023) that the protein Menin declines in specific hypothalamic neurons as mice age, while genetically disabling Menin produced greater hypothalamic inflammation, thinner skin, reduced bone mass, worse cognitive performance, and modestly shortened lifespans.
- Gene delivery to the ventromedial hypothalamus in 20-month-old mice restored Menin levels and reversed skin thinning, bone loss, and impairments in learning, memory, and balance within 30 days, with treated mice also outliving untreated controls in a survival analysis.
- Menin controls production of the enzyme PHGDH, which drives synthesis of the amino acid D-serine; when Menin fell, D-serine concentrations in the hippocampus dropped as well, disrupting NMDA-receptor signaling critical for learning and memory.
- D-serine supplementation in older mice's drinking water for three weeks improved learning and memory tests without directly restoring Menin, but it did not reverse age-related losses in bone mass or skin thickness—suggesting Menin influences aging through multiple mechanisms beyond D-serine.
- Subsequent 2024–2026 studies in Cell Metabolism, Nature, and Cell have reinforced the hypothalamus's role as an aging regulator, identifying additional circuits linking hypothalamic neurons to fat tissue, skeletal muscle, daily biological rhythms, and DNA-methylation-based biological age in mice.
Why it matters: This research identifies the hypothalamus as a potential command center for whole-body aging, with the Menin pathway offering two intervention routes: gene therapy that reversed multiple age-related changes in mice, or D-serine supplementation that improved memory but not physical decline. That split suggests any eventual human therapy would be targeted, not a blanket anti-aging treatment.
